I am a PhD student in the lab. I am looking at what effects does the Origin of Replication have on global expression and chromosomal structure in E.coli. Earlier studies have shown an effect of the chromosomal position of gene on its expression by inserting reporter genes on different positions of the E.coli chromosome. This can be due to various local and global reasons like the local supercoiling, effect of global regulators on that region of the chromosome and, the gene dosage gradient. I am moving the origin of replication in different regions of the chromosome to look at global changes and perturbations.
